Sex differences in fear memory consolidation via Tac2 signaling in mice

چکیده
Abstract Memory formation is key for brain functioning. Uncovering the memory mechanisms helping us to better understand neural processes in health and disease. Moreover, more specific treatments fear-related disorders such as posttraumatic stress disorder phobias may help decrease their negative impact on mental health. In this line, Tachykinin 2 (Tac2) pathway central amygdala (CeA) has been shown be sufficient necessary modulation of fear consolidation. CeA-Tac2 antagonism its pharmacogenetic temporal inhibition impair male mice. Surprisingly, we demonstrate here opposite effect Tac2 blockade enhancing consolidation females. Furthermore, show that CeA-testosterone males, CeA-estradiol females Akt/GSK3?/?-Catenin signaling both mediate opposite-sex differential regulation memory.

A Role for Tac2, NkB, and Nk3 Receptor in Normal and Dysregulated Fear Memory Consolidation
The centromedial amygdala (CeM), a subdivision of the central amygdala (CeA), is believed to be the main output station of the amygdala for fear expression. We provide evidence that the Tac2 gene, expressed by neurons specifically within the CeM, is required for modulating fear memories. Tac2 is colocalized with GAD65 and CaMKIIα but not with PKCd and Enk neurons in the CeM. Moreover, the Tac2 ...
متن کاملCerebellar vermis H2 receptors mediate fear memory consolidation in mice
Histaminergic fibers are present in the molecular and granular layers of the cerebellum and have a high density in the vermis and flocullus. Evidence supports that the cerebellar histaminergic system is involved in memory consolidation. Our recent study showed that histamine injections facilitate the retention of an inhibitory avoidance task, which was abolished by pretreatment with an H2 recep...
متن کاملPKMζ contributes in consolidation, retrieval and maintenance of amygdala dependent fear memory in rats
Introduction: Protein kinase M zeta (PKMζ) is assumed to be actively involved in retainig long-term potentiation. The goal of this study was to investigate the role of PKMζ in basolateral amygdala (BLA) upon acquisition, consolidation, retention and retrieval of memory using a specific inhibitor of PKMζ. Methods: Sixty male wistar rats underwent stereotaxic surgery and were cannu...
متن کاملAmygdala-Dependent Fear Memory Consolidation via miR-34a and Notch Signaling
UNLABELLED Using an array-based approach after auditory fear conditioning and microRNA (miRNA) sponge-mediated inhibition, we identified a role for miR-34a within the basolateral amygdala (BLA) in fear memory consolidation. Luciferase assays and bioinformatics suggested the Notch pathway as a target of miR-34a. mRNA and protein levels of Notch receptors and ligands are downregulated in a time- ...
